چکیده

This research was designed to assess the inhibitory action of aliskiren, L-carnitine, and their combined treatment on JAK2/STAT3/SOCS3 signaling pathway in cardiac hypertrophy induced by isoproterenol injection rats. Wistar rats were injected with (5 mg/kg/day) for 15 days induction hypertrophy. Hypertrophied animals concurrently treated daily aliskiren (50 mg/kg) and/or L-carnitine (200 mg/kg). Either or significantly reduced elevated relative heart weight a concomitant reduction brain natriuretic peptide, creatine kinase-MB, troponin T animals. Additionally, myocardial interleukin-6, lipid peroxidation, markedly increased glutathione content. Aliskiren also attenuated fibrosis as evidenced significant decrease collagen I transforming growth factor-β1. The biochemical results further confirmed improvement histopathological architecture. Interestingly, down-regulated expression JAK2, STAT3, SOCS3 hypertrophied Conclusively, aliskiren/L-carnitine regimen may ameliorate through mitigating oxidative stress, inflammation, IL-6/JAK2/STAT3/SOCS3 pathway.

Selumetinib, an Oral Anti-Neoplastic Drug, May Attenuate Cardiac Hypertrophy via Targeting the ERK Pathway

AIMS Although extracellular-regulated kinases (ERK) are a well-known central mediator in cardiac hypertrophy, no clinically available ERK antagonist has been tested for preventing cardiac hypertrophy. Selumetinib is a novel oral MEK inhibitor that is currently under Phase II and Phase III clinical investigation for advanced solid tumors. In this study, we investigated whether Selumetinib could ...

Hydrogen (H2) Inhibits Isoproterenol-Induced Cardiac Hypertrophy via Antioxidative Pathways

Background and Purpose: Hydrogen (H2) has been shown to have a strong antioxidant effect on preventing oxidative stress-related diseases. The goal of the present study is to determine the pharmacodynamics of H2 in a model of isoproterenol (ISO)-induced cardiac hypertrophy. Methods: Mice (C57BL/6J; 8-10 weeks of age) were randomly assigned to four groups: Control group (n = 10), ISO group (n = 1...

Rapamycin Attenuated Cardiac Hypertrophy Induced by Isoproterenol and Maintained Energy Homeostasis via Inhibiting NF-κB Activation

Rapamycin, also known as sirolimus, is an immunosuppressant drug used to prevent rejection organ (especially kidney) transplantation. However, little is known about the role of Rapa in cardiac hypertrophy induced by isoproterenol and its underlying mechanism. In this study, Rapa was administrated intraperitoneally for one week after the rat model of cardiac hypertrophy induced by isoproterenol ...

Diallyl disulfide ameliorates isoproterenol induced cardiac hypertrophy activating mitochondrial biogenesis via eNOS-Nrf2-Tfam pathway in rats

The beneficial effect of garlic on cardiovascular disease is well known. However, the use of raw garlic against cardiac hypertrophy is not established. In the present study we explored whether raw garlic and its compound, diallyl disulfide (DADS) could inhibit hypertrophy through H2S and/or mitochondrial biogenesis. Cardiac hypertrophy was induced in rat by giving isoproterenol at the dose of 5...

Differential regulation of proteasome function in isoproterenol-induced cardiac hypertrophy.

RATIONALE Proteasomal degradation is altered in many disease phenotypes including cardiac hypertrophy, a prevalent condition leading to heart failure. Our recent investigations identified heterogeneous subpopulations of proteasome complexes in the heart and implicated multiple mechanisms for their regulation. OBJECTIVE The study aimed at identification of molecular mechanisms changing proteas...
